Target
Function
Serpin A12 (OL-64, Vaspin, Visceral adipose-specific serpin, Ser A12) is a glycoprotein that is a class A member of the serine protease inhibitor (serpin) family. In humans, Serpin A12 is encoded by the SERPINA12 gene. First discovered in 2005, Serpin A12 was highly expressed in white adipose tissue of Otsuka Long Evans Tokushima Fatty Rats at the same time that the rats' obesity and insulin plasma levels reached a peak, at around 30 weeks old. Eventually, it was found to be expressed in visceral and subcutaneous adipose tissue of obese humans, leading the protein to be linked with obesity, glucose metabolism, and insulin resistance.

Sandwich ELISA analysis of Human Serpin A12 matched pair antibodies
Capture: HA725480, Human Serpin A12 Rabbit mAb [PSH24-20] Detector: HA725481, Human Serpin A12 Rabbit mAb [PSH24-21]
Elisa assay was performed by coating wells of a 96-well plate with 100 µl per well of capture antibody (HA725480) diluted in carbonate/bicarbonate buffer, at a concentration of 2 μg/ml overnight at 4℃. Wells of the plate were washed, blocked with 150 µl 0.05% tween-20 1% BSA blocking buffer, and incubated with serial diluted Recombinant Human Serpin A12 protein (HA211482) starting from 4,000 pg/ml to 0 pg/ml and detect antibody (HA725481, Biotin, 0.2 µg/ml) for 1 hour at 30℃ with shaking. Then the plate was washed and incubated with 100 µl per well of SA-HRP for 0.5 hour at 30℃ with shaking. Detection was performed using an Ultra TMB Substrate for 10 minutes at room temperature in the dark. The reaction was stopped with sulfuric acid and absorbances were read on a spectrophotometer at 450 nm.
Interpolated concentrations of native Serpin A12 in human serum samples.
Capture: HA725480, Human Serpin A12 Rabbit mAb [PSH24-20] Detector: HA725481, Human Serpin A12 Rabbit mAb [PSH24-21]
The concentrations of Serpin A12 were measured in duplicates, interpolated from the Serpin A12 standard curve and corrected for sample dilution. Undiluted samples are human serum 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Serpin A12 concentration was determined to be 1,548 pg/ml in human serum.
